Anything you do to remove roughness and sharp contours will help.
Definitely open up the bores if you're going to a v8 TB, there is a lot of
work that can be done under the TB bores as well. That area is generally
pretty sloppy.

Smoothing the runners, making their edges more rounded and smooth will have
more of an effect than smoothing the plenum walls. Once you get the
bellypan off and stare at the underside of the intake things will make
sense.

Step back and think of how the air flows through the manifold, those areas
where intake velocity will be higher (ends of runners) will require more
attention to smooth transitions (no sharp corners or edges!)
